ABSTRACTThe efficacy and safety of doxycycline (doxydine) as an antibiotic were evaluated in a randomized double-blind, placebo-controlled study conducted in a tertiary care university hospital. The primary outcome was the change in the oral antibiotic efficacy. Secondary outcomes were the incidence of adverse events and number of events (AEs) on the laboratory tests. A total of 675 patients were randomized to receive either the study drug or a placebo in a clinical trial and the results were analyzed using logistic regression analysis. The primary end point was the change in the oral antibiotic efficacy. The incidence of AEs was significantly increased by doxydine with a significant increase from the placebo group (3.3% vs 1.2% and 2.1% vs 0.9%, respectively; P<0.0001). The incidence of AEs on the laboratory tests was significantly increased in the doxydine group (4.2% vs 2.1%; P<0.0001). The incidence of adverse events was significantly decreased in the doxydine group compared to the placebo group (3.3% vs 1.2%; P=0.01). The incidence of AEs on the laboratory tests was significantly decreased in the doxydine group compared to the placebo group (3.3% vs 1.2%; P=0.0001). The incidence of AEs on the laboratory tests was significantly decreased in the doxydine group compared to the placebo group (2.2% vs 1.2%; P=0.0004). The incidence of AEs on the fecal-oral test was significantly increased in the doxydine group (3.3% vs 1.2%; P=0.0001). The incidence of adverse events was decreased in the doxydine group compared to the placebo group (3.3% vs 1.2%; P=0.0001).
